If you are getting this error message for a host named site collection even after assigning appropriate permissions, do check that the SharePoint Web Application and host named site collection are pointing to the same IP address! You can quickly check if it is a DNS issue by adding the DNS aliases to the host file at C:\Windows\System32\drivers\etc\hosts, starting a command prompt and typing the command “ipconfig /flushdns”, closing all the browser window, and navigating to your site again.
